SUBSTANCE: described is a method for sequencing a target nucleic acid on a matrix, involving: (a) obtaining a first and a second population of granules, wherein: the first population of granules contains oligonucleotides containing first capture probes, first barcodes and barcode primer binding sites adjacent to the first barcodes, and the second population of granules comprises oligonucleotides containing second capture probes, second barcodes and barcode primer binding sites adjacent to the second barcodes; (b) obtaining a first and a second plurality of polynucleotides, wherein: the first plurality of polynucleotides comprises first target nucleic acids, wherein the first plurality of polynucleotides is in solution, and the second plurality of polynucleotides comprises second target nucleic acids, wherein the second plurality of polynucleotides is in solution; (c) hybridising the first target nucleic acids with the first capture probes to obtain hybridised first granules and hybridising the second target nucleic acids with the second capture probes to obtain hybridised second granules; (d) random distribution of hybridized first granules and hybridized second granules on matrix; (e) decoding the positions of the first and second beads on the array by sequencing the first and second bar codes on the array; and (f) extending the first and second capture probes to obtain nucleotide sequence data for the first and second target nucleic acids.
EFFECT: invention provides accurate genotyping.
