SUBSTANCE: described is a nucleic acid library for analysing DNA and RNA from a sample, created simultaneously from RNA and DNA in one compartment, containing a library of second strand complementary DNA (cDNA) molecules, obtained from mRNA molecules and containing nucleic acids having a first label, and a genomic DNA (gDNA) library obtained from genomic DNA tagged with a second label and 3' poly-A tail using transosomes with poly-A transposons, and wherein said cDNA and gDNA libraries are jointly compartmentalized and obtained in the same medium. Also described is a flow cell device for simultaneous production of RNA and DNA libraries in one compartment, comprising a capture probe based on poly-T, configured to hybridise with 3' poly-A tail of RNA, wherein the capture probe contains a first common sequence (CS1); a poly-T capture probe configured to hybridise with 3' poly-A DNA tail, wherein the capture probe comprises CS1; wherein each capture probe is configured to simultaneously analyse RNA and DNA from the sample in one compartment. Disclosed is a method for simultaneous analysis of DNA and RNA from a sample in one compartment, involving: providing a sample comprising DNA and RNA, wherein RNA comprises 3' poly-A tail; DNA tagmentation with 3' poly-A tail using transosomes with poly-A transposons; bringing a sample in one compartment into contact with a poly-T capture probe configured to hybridise with 3' poly-A tail of RNA and DNA, wherein the capture probe comprises a first common sequence (CS1); hybridisation of the poly-T-based capture probe with RNA and with DNA, thus simultaneously capturing DNA and RNA; and DNA and RNA analysis. Also disclosed is a method for simultaneous creation in one compartment of a nucleic acid library containing gDNA and cDNA, comprising: providing a sample containing DNA and RNA, wherein RNA contains 3' poly-A tail; DNA 3' poly-A tagmentation with the use of transosomes with poly-A transposons; bringing a sample in one compartment into contact with a poly-T capture probe configured to hybridise with 3' poly-A tail of RNA and DNA, wherein the capture probe comprises a first common sequence (CS1); hybridisation of the capture probe with RNA and DNA, thus simultaneously capturing DNA and RNA; converting RNA into a first and/or second strand of cDNA; and the simultaneous creation of a cDNA library and a gDNA library from hybridised RNA and DNA. Described is a kit for simultaneous analysis of DNA and RNA from a sample in the same compartment, comprising transposition reagents and a poly-T-based capture probe configured for hybridisation with 3' poly-A tail of RNA and 3' poly-A tail of the DNA, wherein said capture probe comprises a first common sequence (CS1).
