FIELD: chemistry.
SUBSTANCE: invention relates to production of functional polymer materials, namely, is a carrier for use in solid-phase synthesis of oligonucleotides. Carrier for solid-phase synthesis is described by formula:
where R1= -(CH2)n-, n is integer from 0 to 3, or R1 = -O-(CH2)m-O-, m is integer from 1 to 4; and R2= -OH or -NH2; wherein the preparation of the support for solid-phase synthesis includes the following steps: first, aqueous and oil phases are obtained; aqueous phase contains water, a dispersing agent and an inorganic salt; oil phase contains: a cross-linking monomer, a monovinyl compound, a functional monomer, a modified monomer; pore-forming agent and initiator. Oil phase is then added to the aqueous phase while stirring and heating to carry out a polymerisation reaction, after which the pore-forming agent is removed. As a result, a porous polymer resin is obtained, which is capable of undergoing further reaction to obtain a solid-phase synthesis support containing a hydroxyl group and an amino group as functional groups. Further reaction includes obtaining a support for solid-phase synthesis, containing an amino group as a functional group through Gabriel reaction. Obtained support has content of hydroxyl groups or amino groups from 100 to 1000 mcmol/g; particle size in range of 35-200 mcm and average pore diameter of 10-200 nm.
EFFECT: obtaining oligonucleotides using the disclosed solid-phase synthesis carrier increases output and purity of oligonucleotides.
