FIELD: molecular biology, biotechnology. SUBSTANCE: invention relates to compositions (K) used for polymerization immobilization of biological macromolecules in hydrogels on formation of biochips K = aA+bB+cC+dD+eE involving: A is a monomer based on derivatives of acrylic and methacrylic acids; B is a water-soluble cross- linking agent; C is a modified biological macromolecule containing unsaturated group; D is a water-soluble compound as a component of medium where copolymerization reaction is carried out; E means water where a, b, c, d, e mean percent content (X) of each component in the composition (X = m/v x 100% for solid substances and X = v/v x 100% for liquid substances) where total content of monomer and cross-linking agent is in the range 3-40% (3 ≤ (a + b) ≤ 40), ratio of monomer to cross-linking agent is in the range (97:3)- (60:40) and percent content of components C, D and E is in the range 0.0001% ≤ c ≤ 10%; 0% ≤ d ≤ 90%; 5% ≤ e ≤ 95%. Invention relates also to method of their preparing, modified biologically important compounds DNA and proteins, biochip where silt layer formed on backing is separated for empty spaces for some cells being each cell can comprise or no comprise immobilized macromolecules and macromolecules immobilized in different cells can be differed by their nature and properties. Invention relates also to methods for carrying out polymerase chain reaction on biochip. EFFECT: improved preparing and carrying out method. 34 cl, 2 tbl, 8 dwg
