FIELD: oil and fat industry.

SUBSTANCE: method for determining content of squalene in vegetable oil and products of processing oil-bearing raw materials involves preparing a reference sample by mixing a standard sample of squalene and a solvent, analysis of a reference sample by chromatography-mass spectrometry, plotting a calibration curve of the dependence of peak height on squalene concentration and a regression equation, preparation of the analysed sample by mixing a weighed portion of the analysed sample with 2 % alcohol solution of potassium hydroxide in a volume ratio of 1:10, followed by holding, diluting with distilled water in a volume ratio of 1:1, cooling to room temperature, six-fold extraction with hexane, taken in amount of 300 cm3, by mixing the extracts obtained after extraction and successive washing with 50% ethyl alcohol pH 8.0 and pH 7.0, evaporating the washed extract in a rotary evaporator to obtain a precipitate, drying at temperature of 80 °C to constant weight and dissolution in hexane at temperature of 40 °C, analysis of the obtained test sample by chromatography-mass spectrometry, quantitative determination of squalene in the test sample by absolute calibration, wherein the analysed sample used is distillation refining of vegetable oil or vegetable oil, mixing of a standard sample of squalene and a solvent, when preparing a reference sample, is carried out in the concentration range from 0.1 mg/ml to 10 ml/mg of squalene, and holding the solution obtained by mixing a weighed portion of the analysed sample with 2% alcohol solution of potassium hydroxide is carried out for 90 minutes, wherein quantitative determination of squalene in analysed sample is carried out by formula y=[1.603∙10-3]∙x+[2.332∙10-2], (1), where y is squalene content, wt.%; x is peak area.

EFFECT: invention enables to determine content of squalene with an error of not more than ±0.009 %, as well as to expand the objects of study, in particular, in addition to distillation refining of vegetable oil, to use vegetable oils.
