FIELD: medicine.
SUBSTANCE: invention refers to medicine, namely to a method for preparing an autologous bone marrow aspirate concentrate. Method for producing an autologous bone marrow aspirate is characterized by that the bone marrow aspiration procedure is performed under aseptic conditions under local or combined anaesthesia from an upper posterior iliac spine, after palpation of bone landmarks of posterior iliac crest and sacroiliac joint, performing infiltration of soft tissues and periosteum of superior posterior iliac crest with 0.5% solution of ropivacaine 20–25 ml, then with a needle scalpel, an incision of the skin with length of 0.3–0.4 cm is made, through which a trepanobiopsy needle is inserted, pre-washed with a heparin solution, having passed the cortical layer at depth of 2–2.5 cm, the mandrin is removed, connecting a syringe pre-filled as an anticoagulant with a heparin solution of 500 units per 1 ml of bone marrow, followed by aspiration of bone marrow in volume of 80–200 ml, the obtained bone marrow is placed in test tubes with volume of 50 ml, then centrifuged at 400g for 2 minutes from 2 to 6 times with visual control of material separation into fractions with erythrocyte sedimentation to 25–30 ml, after the bone marrow is divided into fractions, 20–25 ml erythrocyte sediment is removed from the bottom of each test tube with a syringe with long 18G needle, and the remaining material is centrifuged for 10–15 minutes at 700–1,000g, using a syringe with a long needle, from each test tube removing the surface layer of plasma, preventing aspiration of the sediment, remaining concentrate of bone marrow aspirate from test tubes is collected by means of long 18G needle into two 10-ml syringes in volume of 5–7 ml into each syringe for further introduction into necrosis centre.
EFFECT: method described above enables to obtain a concentrate of the bone marrow aspirate with an increased number of bone marrow cells and reduce the time for preparing the concentrate of the bone marrow aspirate.
