SUBSTANCE: invention relates to protease-activated polypeptides and can be used for their recombinant production. Disclosed is a target tissue-specific polypeptide comprising an antigen-binding domain, a cleavage site and a transporting fragment, wherein the transporting moiety comprises a constant region and an inhibiting domain which inhibits the antigen-binding activity of the antibody, and wherein the antigen-binding domain has a shorter blood half-life compared to the transport moiety, wherein the cleavage site comprises a protease cleavable sequence that is cleaved by a target tissue-specific protease, in which the cleavage site is located near the boundary between the antigen-binding domain and the constant region of the antibody.
EFFECT: invention provides obtaining a target tissue-specific polypeptide, in which a cleavage site is cleaved so that the antigen-binding domain can be released from the polypeptide and/or association of inhibiting domain and antigen-binding domain is degraded, in which the released antigen-binding domain has higher antigen-binding activity than before release.
