SUBSTANCE: disclosed is a method for preparing samples for amino acid analysis under conditions of reversed-phase high-performance liquid chromatography, involving obtaining an amino acid hydrolysate using 6 M hydrochloric acid solution, derivatization with phenyl isothiocyanate using a chromatographic column, where hydrolysis of a sample of protein or peptide nature is carried out with microwave irradiation at temperature of 150-160 °C for 25-55 minutes with a microwave radiation power of 1,800 W at frequency of 2,450 MHz and pressure of 2.8-3.5 atm, the hydrolysate is dried in a vacuum, sodium hydroxide solution 1 M is added to pH=7.5-7.8, mixed, derivatization is carried out for 30 minutes at temperature of 25 °C with a solution of phenyl isothiocyanate in a mixture of isopropyl alcohol-dimethylsulfoxide, taken in a volume ratio of 1:1.
EFFECT: invention increases sensitivity of the derivatizing reagent while simplifying sample preparation.
