SUBSTANCE: invention relates to biotechnology and microbiology, and can be used for controlled synthesis of secreted stress proteins of Francisella tularensis. Disclosed is a method of producing secreted stress proteins of Francisella tularensis, involving stress exposure on a bacterial culture when cultivated in a liquid nutrient medium by adding 38% H2O2 to a final concentration of 5 mM and increasing temperature to 42 °C for 2 hours, sterilizing filtration of the culture liquid, precipitation of the protein fraction with ammonium sulphate at 55% to obtain Bfr proteins, at 70% to obtain GroEL/GroES proteins, followed by purification by gel chromatography and ion-exchange chromatography.
EFFECT: method enables increasing synthesis and secretion of stress proteins Bfr and GroEl/GroEs Francisella tularensis with subsequent recovery of these antigens in a biologically active state from the culture fluid without impurities of other components of the bacterial cell.
