METHOD FOR DIAGNOSING CHRONIC ENDOMETRITIS OF VARIOUS SEVERITY IN PATIENTS WITH ABNORMAL UTERINE BLEEDING, WITH NO HISTORY OF ENDOMETRIOPATHY, BY IMMUNOHISTOCHEMICAL STUDY OF CELLULAR IMMUNITY DISORDER IN ENDOMETRIUM Russian patent published in 2024 - IPC G01N33/53 A61B5/00

Abstract RU 2830413 C1

FIELD: medicine.

SUBSTANCE: invention relates to a method for determining a degree of manifestation of chronic endometritis (CE). Endometrial biopsy or hysteroscopy, diagnostic curettage of the uterine mucosa in the middle stage of the proliferation phase—7–11 days of 28-day menstrual cycle in a spontaneous cycle. Pipeline endometrial biopsy is performed using a disposable aspiration probe. Obtained material is embedded in paraffin after histological preparation in an automatic histoprocessor; then, sections with thickness of 4–6 mcm are stained with hematoxylin and eosin and histochemical stain according to Mallory. Immunohistochemical staining is performed on dewaxed and dehydrated sections using avidin-biotin immunoperoxidase method. Expression levels of CD138+, CD56+, CD20+, CD4+, CD8+ in the endometrium are determined. Percentage of positively stained cells is counted with assignment of points, and immunohistochemical reaction is assessed. Further, the consistency of cell immunity is assessed by the ratio of CD4+/CD8+ markers in the endometrium and the presence of fibrosis with assignment of points. Sum of the obtained points is calculated and the severity of CE is determined: at 15 and more points—pronounced CE; at 9–14 points—moderately expressed CE; at 5–8 points—weak CE, at 0–4 points—CE is unlikely.

EFFECT: invention provides high sensitivity of verifying cellular immunity and determining the severity of CE, allowing to predict a positive outcome of the onset and bearing of pregnancy, in patients with a history of abnormal uterine bleeding and menstrual irregularities.
