FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, particularly to a method for minimally invasive recovery of bacterial DNA from a biopsy material of prostate tissue. Sampling of prostate tissue biopsy material is performed by prostate biopsy, after which the sample is subjected to dry cryopreservation, after which the intact DNA is purified from the prostate tissue, 160 mcl TE buffer and 20 mcl lysozyme are added to the prostate tissue sample, thoroughly mixed, shaken, incubated at 37 °C, 20 mcl Proteinase K solution and 220 mcl Lysis buffer are added, shaken, incubated again at 56 °C, 20 mcl of RNase A solution is added, mixed on a vortex, followed by incubation at 56 °C, 270 mcl of binding buffer is immediately added, shaking and centrifuging, placing the columns inside the sample collection tube and transferring the liquid to the column membrane, centrifuging and adding 500 mcl of the washing solution, the liquid is centrifuged and separated, the column is transferred into a clean microcentrifuge test tube, 50 mcl of water is added, centrifuged and the obtained eluate of bacterial DNA of prostate tissue is cryopreserved for further analysis.
EFFECT: method enables reducing labour intensity, improving accuracy of diagnosing chronic inflammatory diseases of the prostate, reproductive potential, as well as validity of the treatment algorithm selection program.
