FIELD: digital microscopy.

SUBSTANCE: method of autofocusing when digitizing a microscopic preparation under conditions of delay in obtaining coordinates involves continuous scanning of the focusing area, wherein scanning is carried out in 3 steps. First stage includes moving the object table with the sample along the Z axis at maximum speed towards the microscope lens with continuous calculation of the sharpness level Q of the obtained images with the determination of its maximum value and maintaining the current coordinates, when the sharpness level Q exceeds the specified one, the condition of stopping by the sharpness threshold is triggered and the object table, having travelled another distance, stops. Thereafter, a transition to the second stage is performed, which includes moving the object table in the direction from the lens to set the object table at a distance corresponding to the maximum sharpness value determined at the first stage, and maintaining the current sharpness level Q1. Then, the transition to the third stage is performed, which includes moving the object table in the direction towards the lens to set the object table at a distance corresponding to the point of best focus, wherein this stage is repeated by repeated movements of the object table with simultaneous calculation of the sharpness level Q until reaching the point of the best focus, after which the device is considered to be focused.

EFFECT: high speed and stability of autofocusing in conditions of delay in obtaining actual coordinates of the focusing device.
