FIELD: imaging technology.

SUBSTANCE: group of inventions relates to a method for determining and maintaining optimal image focusing quality and to a system for imaging fluorescence objects in a flow cell. The inventions can be used in devices for mass parallel DNA sequencing by synthesis. The method involves determining the quality parameter of focusing coordinates along the Z axis at the optimal value of the image focusing quality parameter for four or more fields of view having the coordinates of reference points along the mutually perpendicular X and Y axes; approximation of the dependence of the Z coordinate on the coordinates of the reference points X and Y by continuous functions and control of the device for moving the micro lens along the Z axis for all positions along the X, Y axes in accordance with the obtained functions of the dependence of the Z coordinate on the coordinates of the reference points X and Y. The system contains a sample platform with a flow cell, a radiation source, an optical unit, a controller with devices for moving the sample platform and a device for moving a micro lens, a focus quality parameter determination unit, an image focus quality optimization unit and a hardware module associated with a controller (6) and an image focus quality optimization unit (11), or controller software module (6), which determine the functional dependence of the coordinates of the micro lens (5) on the Z axis on the coordinates of the platform (1) for samples on the X and Y axes to maintain optimal image focusing quality and set the position of the micro lens on the Z axis for all positions of the platform (1) for samples on the X and Y axes.

EFFECT: implementation of the group of inventions simplifies the imaging system of fluorescence objects of the analyzed DNA samples, and also accelerates the production of a variety of images with optimal focusing quality.
