FIELD: biotechnology.
SUBSTANCE: group of inventions relates to biotechnology, namely genetic engineering. Described is a recombinant protein of receptor-binding domain (RBD) – a structural domain of the spike glycoprotein SARS-CoV-2. Disclosed is a method of producing said protein. Recombinant RBD S-protein of the coronavirus SARS-CoV-2 has size of 437 amino acids with a molecular weight of 48.61 kDa. This protein is obtained by transformation of cells of the strain of methylotrophic yeast Pichia pastoris SuperMan 5-23 with a plasmid synthesized in Escherichia coli cells, containing a sequence in accordance with the sequence RBD Cov19: MASNFRAQPTESIVRFPNITNLCPFGEVFNATRFASVYAWNRKRISNCVADYSVLYNSASFSTFKCYGVSPTKLNDLCFTNVYADSFVIRGDEVRQIAPGQTGKIADYNYKLPDDFTGCVIAWNSNNLDSKVGGNYNYLYRLFRKSNLKPFERDISTEIYQAGSTPCNGVEGFNCYFPLQSYGFQPTNGVGYQPYRVVVLSFELLHAPATVCGPKKSTNLVKNKCVNFNFNGLTGTGVLAESNKKFLLFQQFGRDIADTTDAVRDPQTLEILDITPCSFGGVSVITPGTNTSNQVAVLYQGVNCTEVPVAIHADQLTPTWRVYSTGSNVFQTRAGCLIGAEHVNNSYECDIPIGAGICASYQTQTNSPRRARSVASQSIIAYTMSLGAENSVAYSNNSIAIPTNFTISVTTEILPVSMTKTSVDCTMYICGHHHHHH, their cultivation at temperature of 30 °C for 24 hours in BMGY medium with composition: 1% yeast extract, 2% peptone, 100 mM potassium phosphate buffer with pH=6.0, 1.34% YNB, 4×10-5% biotin, 1% glycerol and 100 mcg/ml kanamycin and separated by affinity chromatography.
EFFECT: invention can be used in creating ELISA test systems and for designing vaccines against coronavirus infection – COVID-19.
