FIELD: biotechnology.
SUBSTANCE: invention relates to biotechnology, in particular to versions of a composition for inducing an immune response for therapeutic or preventive treatment of a disease. Said composition comprises an effective amount of an RNA polynucleotide comprising a payload coding sequence, wherein 5'-end of the RNA polynucleotide contains m7(3’OMeG)(5')ppp(5')(2'OMeA1)pG2pA3pA4pN5, where N5 is uridine, or modified uridine, or m7(3'OMeG)(5')ppp(5')(2'OMeA1)pG2pA3pN4pN5, where each of N4 and N5 is independently selected from A, G, C, U or modified uridine. Present invention also relates to versions of a pharmaceutical composition in which said RNA polynucleotide is in form of a particle.
EFFECT: present invention provides RNA cassettes with an improved level of expression of the encoded payload and/or increased duration of expression of the encoded payload in vivo.
