FIELD: biotechnology.
SUBSTANCE: method of neutralizing an antigen is provided, which comprises replacing at least one amino acid of the variable region of the antibody with histidine, wherein histidine is substituted with at least one amino acid at a position selected from the group consisting of H27, H31, H32, H33, H35, H50, H58, H59, H61, H62, H63, H64, H65, H99, H100b and H102 of a heavy chain variable region according to Kabat numbering and L24, L27, L28, L32, L53, L54, L56, L90, L92 and L94 in the variable region of the light chain according to Kabat numbering, and where this replacement with histidine increases the value KD(pH 5.8)/KD(pH 7.4), defined as the ratio between KD for the antigen at pH 5.8 and KD for the antigen at pH 7.4, compared with value KD(pH 5.8)/KD(pH 7.4) before replacement with histidine, and where said value of KD(pH 5.8)/KD(pH 7.4) after replacement of histidine is 2 or higher, and (b) in vivo administration of said antibody containing substitution, as determined in step (a), thereby neutralizing the antigen.
EFFECT: invention provides a novel method of neutralizing the antigen.
