METHOD OF CYTOCHROME C PREPARING Russian patent published in 1994 - IPC

FIELD: medicine, biochemistry. SUBSTANCE: 10 kg frozen seal heart are thawed, washed with water, fat and ligaments are removed, and tissue is minced. Mince is extracted with trichloroacetic acid solution (at pH 3.8-4.1) for 2 h, extract is separated by filtration, and NaOH is added up to pH 6.0. Proteins are precipitated with crystalline ammonium sulfate. After filtration crystalline ammonium sulfate is added to filtrate obtained. After settling the solution is filtered, and 20% solution of trichloroacetic acid is added to filtrate. Precipitate is separated by centrifugation, treated with ammonium sulfate solution and centrifuged again. Precipitate of cytochrome c is dissolved in distilled water and frozen at (-10)-(-15) C following by thawing and filtration. Concentrated solution is dialyzed, prepared solution is heated and filtered. Filtrate is diluted with saline solution, filtered under sterile conditions, bottled and lyophilized.
