METHOD OF INSERTION OF HETERODUPLEX DNA INTO GENOME OF ADENOVIRUS CELO AND RECOMBINANT ADENOVIRAL VECTOR CELO/PUC19 Russian patent published in 1995 - IPC

FIELD: biotechnology. SUBSTANCE: DNA insertion into adenovirus genome is carried out by hydrolysis of adenovirus CELO genome and DNA of prokaryotic vector pUC19 with restriction endonuclease XbaI, and also by hydrolysis of DNA of adenovirus CELO with restriction endonuclease NotI, and by hydrolysis of DNA fragment CELO/XbaI with restriction endonuclease NotI. Then in system in vitro using method of ligation genome of adenovirus CELO is reconstructed successively from DNA adCELO fragments, such as NotI-A, XbaIA-NotI and XbaIE + DNA pUC19XbaI. Using recombinant DNA CELO reconstructed in system in vitro cell transfection of chorion-allantois envelope of chicken embryo is carried out. As a result of transfection with recombinant DNA CELO full-value virions of avian adenovirus CELO were formed which contain heteroduplex DNA in nonessential part of genome - DNA of prokaryotic vector pUC19.
