METHOD OF PREPARING VACCINE AGAINST NECROBACTERIOSIS IN CATTLE Russian patent published in 1995 - IPC

FIELD: veterinary microbiology, biotechnology. SUBSTANCE: epizootic culture of necrobacteriosis pathogen is used for vaccine preparing. Culture is grown, inactivated and separated from cultural fluid. Antigenic substances were extracted from biomass and cultural fluid using methods protecting antigen activity at extraction. For antigen extraction from microbial cells biomass is resuspended in distilled water preparing the suspension at concentration 150-250 billion cells/ml. Antigen is extracted from suspension by successive triple freezing-thawing out and their homogenization. Homogenate is centrifuged at 15000-20000g, and protective antigen is isolated from supernatant. The latter is cytoplasmatic fraction containing endotoxin. Simultaneously, exotoxin is isolated from cultural fluid by its precipitation with gelatin at concentration 0.03-0.06 wt.-% Antigens were combined with oily-lanolin adjuvant at the ratio 1:1.
