METHOD OF PREPARING VACCINE FOR NECROBACTERIOSIS CONTROL IN ANIMALS Russian patent published in 1998 - IPC

FIELD: veterinary microbiology, biotechnology. SUBSTANCE: method involves the use of industrial strain of Bacteroides necrophorum ((ВИЭВ)) selected by character of maximal immunogenic and antigenic activity. Strain is grown and biomass is separated from cultural fluid by centrifugation. Exotoxin is isolated from cultural fluid by ultrafiltration on hollow fibers (pore size is 1.3-1.7 kDa) at protein content 5.5-6.0 mg/ml and inactivated with formalin. Biomass is resuspended in saline solution up to concentration 10-120 billion cells/ml and endotoxin is extracted from prepared suspension by multiple freezing and thawing. Then endotoxin is separated from cellular deitritus by centrifugation and protein concentration is brought about to level 2.5-3.0 mg/ml. Obtained endotoxin and exotoxin are mixed at an equal volume ratio and adjuvant based on mineral oil and lanolin is added. Obtained preparation is used for preparing vaccine used for prophylaxis and treatment of cattle necrobacteriosis. EFFECT: simplified technology, decreased time and energy consumption, providing the lasting and expressing immunity against necrobacteriosis.
