FIELD: technology of protein purification. SUBSTANCE: method involves the use of preparative column chromatography of low pressure. Aqueous solution of insulin-raw (pH 3.0-3.5) is passed through ionite - product of copolymerization of methacrylic, acrylic acid and ethylene glycol dimethacrylate at swelling degree index Ksw = 4.5-4.8 and full exchange capacity 9.2-9.8 mg x eq./g. Process is carried out by passing an aqueous solution of insulin-raw (pH 3.0-3.5) through the indicated ionite which is equilibrated preliminary with 10-16% aqueous solution of isopropanol (pH 3.0-3.5). Then ionite is impregnated with 10-16% aqueous solution of isopropanol (pH 3.0-3.5) and insulin is eluted with 20-25% aqueous solution of isopropanol (pH 3.4-3.9) containing 0.05-0.15 M ammonium acetate. The end product is isolated from eluate by precipitation. EFFECT: increased degree of insulin purification (98.7%).
