FIELD: protein chemistry, medicine, endocrinology, pharmacy. SUBSTANCE: method involves the use of HPLC method for preparing highly purified insulin. Method is based on preparative pericritical reverse-phase HPLC and selective insulin desorption. Eluents: mixtures of aliphatic alcohols with an aqueous 0.05- 0.15 M alkaline metals or ammonium salts. The first eluent has 1-15 vol. % aliphatic alcohol and the second one (desorbing) - 16-30 vol. % aliphatic alcohol. Column regeneration is carried out with the third eluent containing 50-60 vol. % aliphatic alcohol. Output method is 0.28-0.41 g/cm2 x h, insulin content in product is 98.5%, proinsulin content is 8 m. d., deamidoinsulin content is 1.4%. EFFECT: improved method of insulin preparing.
