FIELD: microbiology, engineering enzymology. SUBSTANCE: method involves growing bacterium Propiobacterium freudenreichii ssp. shermanii without additional aeration. Antibiotic bleomycetin is added 2-2.5 h before culturing termination. Cells were separated from medium, resuspended in potassium-phosphate buffer, permeabilized with Triton X-100, frozen in liquid nitrogen, thawed and treated with glutaraldehyde. After dialysis cells were placed in incubation medium containing the definite ribonucleotide and after 2-3 h incubation the corresponding 2'-deoxyribonucleoside-5'-diphosphates were isolated from medium. Yield is 25-37% (by substrate). EFFECT: improved method of preparing, increased yield.
