METHOD AND HOMOGENIZING VALVE FOR PRODUCING LIPOSOMAL FORM OF ALPHA-TOCOPHEROL Russian patent published in 1997 - IPC

FIELD: medicine. SUBSTANCE: according to method, phospholipids are solved in organic solvent with subsequent evaporation, drying by means of rotary evaporator, hydration, homogenization at high pressure in continuous process, and sterilizing filtration of liposomal suspension. Phospholipids and alpha-tocopherol solved in organic solvent are mixed, filtered under pressure in atmosphere of inert gas. Then, this mixture upon hydration is subjected to cyclic homogenizing in closed circuit under pressure of 10-200 MPa at temperature of 32-35 C. With that, ratio of alpha-tocopherol in relation to phospholipids is 1:6 in mass respectively. Used in function of phospholipids is mixture of lecithin and cholesterol taken in any ratio. Used for solving of phospholipids is ethyl alcohol, and for solving of alpha-tocopherol use is made of chloroform. In performing process of homogenizing, number of cycles can be from 1 to 50. Realization of aforesaid method is effected with the help of homogenizing valve having supporting seat with inlet and outlet holes with sharp edge and circular groove. Valve is provided with lock made in the form of cylinder with tapered end surface and supporting seat installed in outlet hole with possible interaction between lock tapered surface and sharp edge of seat. Circular groove is made in internal surface of supporting seat and is located between sharp edge of seat and outlet hole, and ratio of width and depth of circular groove is from 3:2 to 1: 5 respectively. It is feasible that value of solid angle of tapered end surface is from 15 to 165 deg. EFFECT: high efficiency. 4 cl, 3 dwg
