Method involves contact of nucleic acid sample with diagnostic primer that is complementary with respect to diagnostic site of base sequences in target. Method provides spreading the diagnostic primer only on a matrix under the corresponding conditions if terminal nucleotide of diagnostic primer is complementary to either with respect to the proposed variant nucleotide or to the corresponding normal nucleotide of the target base sequence.
