FIELD: biotechnology. SUBSTANCE: fragments of genome DNA encoding resistance to doxorubicin involving identified restriction maps are obtained. Fragments are prepared by cleavage of genome DNA of the strain Streptomyces peucetius DSM-4592 or its related strains with enzyme MboI. Obtained fragments are fractionated to sizes 4-6 kb, ligated with linearized vector pIJ= 702 and the obtained mixture is used for transformation of protoplasts Streptomyces lividans TK-23 that show sensitivity to doxorubicin at concentration 30 mcg/ml, not above. Clones showing resistance to doxorubicin are identified by the growth index in the presence of doxorubicin and DNA-fragment indicated above is isolated from their.
