FIELD: biochemistry, biotechnology. SUBSTANCE: method involves homogenization of crab hepatopancreas in saline buffer solution, fractionation with ammonium sulfate and the following centrifugation. For chromatography method of purification hydrophobic chromatography is used. Enzyme is sorbed on sorbent equilibrated with buffer salt solution at concentration 1.0 M, not less. Enzyme is eluted by step decrease of buffer salt solution concentration to 0.025 M. Polyvinylpyrrolidone carriers are used as sorbent. Ammonium sulfate is used as a buffer solution for product sorption and ammonium hydrocarbonate is used for desorption. Material is fractionated with ammonium sulfate by its multiple reprecipitation with indicated reagent up to 50% saturation. Method provides purification of the end product from enzyme elastase and pigments and ensures to use sorbents that are used many times in technological process of purification of indicated preparation. Invention can be used for preparing the purified preparation collagenase (collase) from animals and the end product can be used in cellular biotechnology, medicine and perfumery.
