FIELD: biotechnology. SUBSTANCE: cotton explant is isolated from germinated seeds and cultured on solid nutrient medium containing glucose for callus formation. When primary callus appears, it is transferred into sucrose-containing solid nutrient medium to be cultured there until secondary callus is developed. Sucrose can likewise be utilized in media as carbon source at the moment when phenol secretions are nearly depleted. As explant, the one isolated from hypocotyl and/or seed-lobe descended from seedling and immature cotton germs is utilized.
