Method involves infection of human diploid cellular culture with measles virus in supporting medium. Human embryo lung fibroblasts - HELF 385/13 are used as a diploid cellular culture. Supporting nutrient medium used for measles virus culturing has the following composition by the first variant, vol.%: medium-199, 40-58; Eagle MEM medium with double content of amino acids, 40-58; aminopeptide, up to 100. By the second variant supporting medium has the following composition, vol. %: medium-199, 40-59; Eagle medium with double content of amino acid, 40-59; arginine, up to 100. Method increases the yield and biological activity of measles virus preparations and provides sufficient purity and stability of the end product.
