FIELD: gene engineering, medicine, vaccines. SUBSTANCE: gene engineering HBs Ag is accumulated in developing chicken embryos which were infected with recombinant pox-virus in allantois cavity or in cultured cells. HBs Ag is isolated and purified by diaphragm filtration on diaphragm cascade at pore size of one of which does not exceed 0.15-0.22 mcm and affinity chromatography on immune sorbent (polyclonal specific antibodies to HBs Ag immobilized on agarose 4B activated by CNBr) and elution with citrate-phosphate buffer (pH 2.0-2.4) and 8M urea (pH 2.0-2.4). Method ensures to obtain vaccine preparation at HBs Ag yield 80-90% and at inert protein content 5-7%. Vaccine is used for hepatitis B prophylaxis and sensitive diagnostica making. EFFECT: improved method of vaccine preparing.
