FIELD: microbiology, molecular genetics. SUBSTANCE: method involves isolation of vaccine strain of Vibrio cholerae which is alternated specifically by technique of recombinant DNA in order to eliminate its virulence without damage of other components that are necessary for immunity. This attenuation is carried out using digestion of plasmids with restriction endonucleases. These plasmids carry the corresponding DNA nucleotide sequences of V. cholerae. Strains without genes encoding toxin or its fragments are obtained by transfer of ligated gene of digested plasmids and the following selection procedures of recombinants with virulent hosts of V. cholerae in vivo.
