SUBSTANCE: invention describes the novel developed plasmids expressing a fused protein where amino acid sequences of lider peptide that is JgG-binding domain of protein A and human proinsulin are bound by arginine residue (plasmid pRproins) or by lysine residue (plasmid pRproins). Human insulin is obtained by the method involving culturing the strain E. coli JM-109-pRproins carrying plasmid pRproins or the strain E. coli JM-109-pRproins carrying plasmid pRproins, isolation of bodies inclusion, solubilization, sulfitolysis and renaturation of fused protein followed by purification of fused protein by ion-exchange chromatorgaphy method, its cleavage with trypsin-like and carboxypeptidase B-like enzymes and isolation of insulin.
