FIELD: chemical, biochemical, physical and other studies. SUBSTANCE: invention is related to processes measuring optical activity of substance in transmitted light. It refers specifically to saccharimetry. Process determining optical activity of substance includes transmission of p-polarized monochromatic light through sample, interaction of light transmitted through sample with standard reflecting structure, achievement of " total suppression " of beam reflected by structure followed by turn of analyzer and computation of angle α of turn of polarization plane caused by sample by formula , where rp, rs are energy reflection factors of structure for p- and s-components at given angle of light incidence; Ao is azimuth of analyzer under " total suppression " of beam before installation of sample; A is azimuth of analyzer under " total suppression " of beam after installation of sample. Light transmitted through sample excites p-polarized surface electromagnetic waves in structure and compensates for phase shift emerging between p- and s-components of reflected beam as result of interaction of light with structure.
