FIELD: medicinal immunology. SUBSTANCE: invention relates to methods of assay of direct effects of different substances, among them drugs, on functional activity of components of complement. Method ensures to assay the effect of substance on complement by inhibition of binding activated component C4 of human complement system based on immunoenzyme method. Method involves firstly sorption of activated subcomponent C1s or complex IgG with component C1 as activating agent of component C4 in micropanel wells. Then human serum taken in the different dilution as a source of component C4 is added to micropanel wells. Human serum has definite concentrations of substance to be tested in experimental series. Human serum has no inhibitor in control seria. Then incubation is carried out. Amount of covalently bound component C4 as result of activation is determined by product of enzymatic reaction using antibodies to C4, namely fraction of immunoglobulins G covalently bound with enzyme. Inhibition constant for test substance in binding activated component C4 is measured by comparison data of amount bound component C4 in the presence and in the absence of inhibitor. Effect of substance on complement is estimated by inhibition constant value. Method ensures to obtain individual inhibition constants.
