FIELD: biochemistry, chemistry of proteins. SUBSTANCE: invention relates to method of synthesis of polypeptids in eucaryotic and procaryotic cell- free systems based on modification of methods of synthesis using cellular lysate or extract in continuous flow regimens or continuous exchange. These regimens include the maintenance of synthesis process by addition of components to reaction mixture that are able to maintain synthesis and removal low-molecular components from reaction mixture that inhibit synthesis. Also, method involves continuous change of concentration of one or some components that are taken among the group Mg2+, K+, NTP, polyamines or their combinations and that determine effectiveness of synthesis in required range of concentrations change. Method does not require the use of expensive exogenous enzyme T7 polymerase. EFFECT: improved method of synthesis, decreased cost of synthesis products. 19 cl, 6 tbl, 6 ex
