FIELD: biotechnology, molecular biology.
SUBSTANCE: invention describes a method for selection of protein using RNA-peptide hybrids. RNA-peptide hybrid comprises RNA molecule that is bound covalently or hybridized by 3'-end of the sequence encoding protein with nucleic sequence able to cease DNA translation. Nucleic acid sequence is joined covalently to protein acceptor and the protein is encoded by covalently bound RNA. Also, invention describes RNA comprising the sequence for translation initiation and start-codon joined functionally with sequence encoding protein. Also, invention describes microchip comprising set of immobilized nucleic acid each of that is hybridized with RNA-protein hybrid. Method for selection of proteins involves the following steps: representing population of RNA molecules with properties enumerated above; translation of sequence encoding protein; selection of RNA-protein hybrid by testing protein for its binding or functional activity. The application of invention allows carrying out the selection of desirable proteins from vat pools and to transform polypeptide sequences in vitro.
EFFECT: improved selection method, valuable biological properties of protein and nucleic acid.
