FIELD: biotechnology, genetic engineering.
SUBSTANCE: invention can be used for preparing secreting human granulocyte modified colony-stimulating factor (hG-CSF). The modified hG-CSF is prepared by replacing the 1-st amino acid in hG-CSF amino sequence of wild type for Ser or the 1-st and the 17-th amino acid for Ser; the 2-d amino acid for Met and the 3-d amino acid for Val, or the 2-d amino acid for Met, the 3-d amino acid for Val and the 17-th amino acid for Ser, or the 17-th amino acid for Thr. Microorganism cells transformed with DNA-containing vector encoding the modified hG-CSF are cultured and hG-CSF is prepared that is secreted into periplasm. Invention provides effective expression and secretion of hG-CSF in microorganism that retains biological activity of wild type protein.
