SUBSTANCE: invention can be used for isolation of proteins with necessary properties from large pools and nucleic acids. Invention involves preparing hybrids "RNA-protein" that involves stage of post-translation incubation with high concentration of monovalent cation. Cations Na⁺, K⁺ or NH+4

are used as a monovalent cation in the concentration from 125 mM to 1.5 M. Method involves also using bivalence and/or the more high-valence cation, for example, Mg²⁺ in the concentration from 25 mM to 200 mM. Method provides preparing library of proteins. Similar method is used for preparing DNA library that involves additionally stage for generating DNA molecule from each RNA-moiety of hybrids. Invention discloses also method for selection of necessary protein and nucleic acid from prepared libraries of proteins and nucleic acids involving preparing and selection of required hybrid "RNA-protein" followed by selection of necessary protein and nucleic acid encoding the necessary protein. Using the invention provides alleviating the isolation of proteins with necessary properties and to solve the problem for isolation and amplification of protein sequence.
