FIELD: biotechnology, molecular biology.
SUBSTANCE: invention can be used for preparing muteins of human IL-4 (interleukin-4) eliciting activity activating T cells and reduced activity activating endothelial cells. Human mutein IL-4 inducing proliferation of T cells and reduced secretion of IL-6 as compared with IL-4 of wild type is prepared by mutation of surface residues in D-spiral of human IL-4 enumerated with accordance to IL-4 of wild type. Muteins IL-4 comprise single, double and triple mutations represented by the following designations: R121A, R121D, R121E, R121F, R121H, R121I, R121K, R121N, R121P, R121T, R121W; Y124A, Y124Q, Y124S, Y124T; Y124/S125A, T13D/R121E; and R121T/E122F/Y124Q in enumeration with accordance to IL-4 of wild type (His = 1). Polynucleotides encoding muteins are cloned to vector that is used for transformation of host-cells followed by culturing. Pharmaceutical compositions containing muteins are used for treatment of patients suffering with state susceptible for treatment with IL-4. Invention provides preparing less toxic mutein IL-4 that gives a possibility for more broad therapeutic using interleukin.
EFFECT: improved treatment method, valuable biological and medicinal properties of mutein.
