SUBSTANCE: invention relates to the strain of bacterium Bacillus stearothermophilus 22 - a producer of site-specific restriction endonuclease recognizing and cleaving the nucleotide sequence 5'-CCNNNN^NNGG-3' and eliciting property of the growth-stimulating activity. Microorganism Bacillus stearothermophilus 22 is a producer of site-specific restriction endonuclease Bst 22I that is isolated from soil of hot springs in peninsula Kamchatka as result of search of producers of site-specific restriction endonuclease. Invention provides preparing site-specific restriction endonuclease recognizing and cleaving the nucleotide sequence 5'-CCNNNNN^NNGG-3'. The yield of enzyme is 50 000 U/g of wet biomass. Site-specific restriction endonuclease Bst 22I is an isoschizomer of restriction endonuclease Bsi YI and can replace the latter in all genetic engineering works. Biomass of cells of the bacterial strain elicits the growth-stimulating activity with respect to seeds of different cultural plants.
