CROSS-LINKED PROTEIN CRYSTAL WITH CONTROLLED DISSOLVING Russian patent published in 2004 - IPC

Abstract RU 2241746 C2

FIELD: chemistry of proteins, biotechnology.

SUBSTANCE: invention relates to cross-linked protein crystals that elicit ability to transform to soluble and active form in medium alteration of environment around indicated crystals. Indicated alteration is taken among group consisting of: alteration of temperature, alteration of pH value, alteration of chemical composition, transfer from concentrate to diluted form, alteration if oxidative-reductive potential of solution, alteration of falling radiation, alteration of the transient metal concentration, alteration of fluorine concentration, alteration of free radicals, alteration of concentration of metallic chelating agents, alteration of shift power acting on crystals or their combinations. In according with one variant of the invention such cross-linked protein crystals are able to release their protein activity with the controlled rate. Method for preparing such cross-linked protein crystals involve selection of conditions necessary for initiating cross-linkage of protein crystals, namely - factors taken among group including: degree of cross-linkage of indicated protein crystal, exposition time duration of indicated protein crystal to a cross-linking agent, amino acid residues taking part in the cross-linking process, the presence of homobifunctional or heterobifunctional properties in cross-linking agent, rate of addition of cross-linking agent to indicated protein crystal, nature of cross-linking agent, chain length of cross-linking agent, surface square of indicated protein crystal, size of indicated protein crystal, form of indicated protein crystal and their combinations. Agent for delivery of protein involves cross-linked protein crystals and agent for its delivery taken among detergent enzymes, cosmetic proteins, pharmaceutical proteins used in agriculture, proteins for vaccines and for treatment against pollutions.

EFFECT: valuable biological properties of cross-linked protein crystals.
