SUBSTANCE: invention relates to methods for sequencing and analysis of nucleic acids. Invention proposes a method for analysis of sequences - specific hybridization. Method involves interaction of target comprising the nucleic acid sequence and a probe comprising the nucleic acid sequence or analog of nucleic acid. Then the first stimulus is applied to the indicated test-sample and signal from the stimulated test-sample is detected. Obtained signal correlates with the affinity binding between target and probe. Then the first signal is calibrated relatively to the first control signal and based on thereof the correspondence degree between target and probe is determined. Then the second stimulus is applied to the test-sample and its signal is detected. The second signal correlates with the affinity binding of target and probe also. Then the second signal is calibrated relatively to the second control signal and based on thereof the correspondence degree of target and probe is determined. Then the quantitative determination of the affinity binding is carried out. The proposed method provides carrying out the pairing degree between target and probe. Electric conductivity and/or fluorescence intensity can be used as signals that correlate with the affinity binding. The proposed method can be used for analysis of specific hybridization between a single-stranded probe and denatured double-stranded target to form triplexes and for duplex hybridization of complexes.
