SUBSTANCE: the present innovation deals with obtaining an antigen out of cattle hepatic tissue due to the following procedures: tissue extraction in sodium chloride isotonic solution, separation of antigen-containing supernatant due to centrifuging, preparation of erythrocytes isolated by centrifuging out of cattle blood due to washing erythrocytic residue in sodium chloride isotonic solution, suspending and keeping erythrocytes in fixative solution followed by tanning and sensitizing of erythrocytes due to mixing with antigen solution with subsequent washing in sodium chloride isotonic solution and resuspending in a conservant up to concentration of ready-to-use product; moreover, extraction of hepatic tissue should be conducted at the ratio of hepatic tissue and sodium chloride isotonic solution being 1:4 for about 18-20 h at 4°C, erythrocytic residue should be washed in phosphate-buffered sodium chloride isotonic solution to be then suspended up to 20% concentration in phosphate-buffer solution containing as a fixative 0.3%-formaldehyde and keep for 4 h at 37°C. Sensitization should be carried out due to mixing one volume of tanned erythrocytes of 50% concentration with 5 volumes of antigen solution and 5 volumes of 0.1%-chromium trichloride solution to be washed in phosphate-buffered sodium chloride isotonic solution at pH being 7.2 and resuspended up to 3% concentration in 0.25%-glycerol solution prepared upon sodium chloride isotonic solution at addition of 0.3%-phenol.
