FIELD: molecular biology, biochemistry.
SUBSTANCE: invention proposes a method for amplification of nucleic acid (NA) involving carrying out a primer reaction elongation on matrix in the presence of at least one nucleotide as a precursor of DNA recognized by a cleaving agent, digestion of elongated primer but retaining a bind with matrix NA by indicated agent to form fragments comprising free OH-group at 3'-end and elongation of prepared fragment on matrix. The distinctive feature of a method is the combining all reactions in a single vessel and using the reaction mixture containing all necessary components, and DNA polymerase, a cleaving agent and the reaction conditions are selected by taking into account their simultaneous functioning. Invention can be used in analytical methods of different designation associated with determination of quantitative and qualitative indices of NA.
EFFECT: improved method for amplification.
