SUBSTANCE: method for conducting a non-isothermal reaction of nucleic acid amplification is disclosed. The method includes mixing of a target sequence with primers, their hybridization to form a duplex containing two gap formation sites located at opposite ends of the duplex or near them, and amplification. In the process of amplification, a gap is created in each gap formation site, gap-containing threads are elongated to form new synthesized nucleic acid and restoration of gap formation sites, and repetition, if necessary, of these stages to obtain multiple copies of new synthesized nucleic acid. A temperature, at which the method is implemented, is decreased in the range of 2-40°C during amplification process, wherein there is no comeback to a temperature, at which a hybridization stage is performed. A device for implementing the specified method is also proposed.
EFFECT: invention provides for an increase in the performance of the amplification method.
