FIELD: medicine.
SUBSTANCE: lysozyme as an activator of complement alternative path is sorbed wells of a microarray. Then a solution containing reagent RB and a tested sample containing human complement factor B of unknown activity is added. It is followed with incubation process with Mg2+ ions added and without Ca2+ ions. Then conjugated enzyme with antibodies against human component "C3" and substratum of this enzyme. The factor B activity is derived from the amount of formed enzymatic reaction product. A kit for assaying functional activity of human complement factor B comprises a flat-bottomed microarray with sorbed lysozyme, reagent RB (human blood serum free from factor B activity), conjugated peroxidase with antibodies to human complement component "C3", substrate buffer and donor blood serum as a reference.
EFFECT: method is characterised with good reproducibility of results and the absence of necessity to use hard-to-reach high-purity preparations of alternative path activators poorly maintaining its activating ability.
