SUBSTANCE: disclosed is a method for expansion of CD34+ cells, involving combined culturing of a substrate suspension in serum-free medium ExVivo 15 with growth factors SCF 50 ng/ml, IL-3 20 ng/ml, IL-6 20 ng/ml, Flt-3 100 ng/ml, wherein the substrate used is OP9 cells, and culturing is carried out at 37°C, 5% CO2 for 12 days and the method of expanding CD34+ involves culturing in a medium of the following composition: medium StemXVivo Serum-Free HSC, containing 100 ng/ml CSF, 100 ng/ml FLT-3, 50 ng/ml TPO, 20 ng/ml IL-3, X-Vivo 15, containing 30 ng/ml IL-3, 50 ng/ml SCF, 10 ng/ml IGF, 50 ng/ml FLT3L, 3 ng/ml GMCSF, 5 ng/ml IL-7, 10 ng/ml SCDF, wherein culturing is carried out at 37°C, 5% CO2 for 14 days, and half of the medium is replaced with fresh medium every three days.
EFFECT: invention enables to increase the number of CD34+ cells fivefold within 12 days or thirtyfold within 14 days, wherein after differentiation, said cells do not exhibit tumoral phenotype.
