FIELD: medicine.
SUBSTANCE: invention describes a method of saliva selenium test involving preparing blood serum, transforming Se(VI) into Se(IV), preparing a selenium-diazol complex, conducting extraction and measuring fluorescence at excitation wave length λexit=366 nm, drawing a calibration curve derived from measuring reference solutions wherein the prepared blood serum 1 ml is added with HCl solution 2.5 ml 0.1N, heated for 30 minutes to 60°C until the volume is 1 ml; thereafter the pre-cooled mixture is added with 2,3-diaminonaphalene (DAN) 2.5 ml , EDTA 1 ml, HCl 50 ml 0.1 N, the prepared mixture is heated at temperature 80°C for 20 minutes; the mixture is cooled to temperature 37°C and extracted in cyclohexane 10 ml; the fluorescence spectrums of the mixture (λreg=480÷600 nm) are measured, and the spectral curves are processed by the use of the Gaussian-Lorentzian curve method.
EFFECT: higher accuracy of saliva selenium test.
