FIELD: medicine.
SUBSTANCE: invention refers to medicine, namely microbiology, clinical and laboratory diagnosis and immunology. A method for preparing hyperimmune rickettsial serums is as follows. Male 10-12-month Chinchilla rabbits 2.5-3.0 kg are immunised at two stages. At the primary immunisation, a mixture of 10% suspension of the rickettsial infectious material (0.5 ml) and Freund's complete adjuvant taken in volume 1.5 times exceeding the amount of the infectious material (0.75 ml) is administered. To stimulate the primary immune response, the mixture injected once in intradermal paravertebral microdoses in 40 points. The infection rickettsial material is homogenised membranes of yolk sacs of chicken embryos infected with R. prowazekii, or homogenised spleen of inbred albino mice 16-18 g infected with rickettsial C.burnetii or R. sibirica. Thereafter 45 day later, provided reducing the antibody titres produced as a result of the primary immunisation, up to 1:20-1:80, the re-immunisation is started: four-fold intravenous injections of the inactivated antigens of the relevant rickettsial pathogens to complement fixation in the concentration of at least 4 antigen units in 1 ml with increasing doses of 1.0, 1.5, 2.0 and 2.5 ml, every 3 days after each injection. For the purpose of the cytokine imbalance correction and additional stimulation of antibody formation at the stage of re-immunisation, recombinant human IL-2 (roncoleukin) in a volume of 1.0 ml at a dose of 250 000 ME is additionally subcutaneously administered. Total blood sampling for preparing serums is performed on the 14th day following the last intravenous injection and specific antibody titres of in indirect immunofluorescent technique not less than 1:5120-1:10240. The length of a complete cycle of immunisation in accordance with the delared method is 68 days.
EFFECT: invention provides reducing the length of immunisation in 2,3 times (from 158 days to 68 days) and preparing the hyperimmune sera, having the collected rickettsial antibody titres in 4-8 times, increasing the material costs for keeping producer animals in 4,7 times and reducing the labour costs in 5,2 times over a production cycle of preparing the rickettsial hyperimmune sera, saving the wages fund in 4,2 times.
