FIELD: biotechnologies.

SUBSTANCE: method includes fixation, washing and dehydration, impregnation in camphene and drying at room temperature. Prior to fixation of the sample the microorganisms were cultivated on the surface of the sterilised cell of the cell package with perforations along the entire diameter of the cell submerged into the nutrient medium. The sample with microorganisms was fixed for 24 hours in the mixture of solutions of paraformaldehyde and glutaraldehyde on a phosphate buffer, pH 7.4, at the temperature of 4°C, and then washed in the phosphate buffer for 20 minutes and in distilled water for 6 minutes. Then the sample was dehydrated in stages in 70% ethyl alcohol for 15 minutes and for 10 minutes in 80%, 96% and 100% ethyl alcohol, impregnated in camphene at t=60°C for 20 minutes. Further samples are dried on air to full sublimation of camphene at room temperature.

EFFECT: prevention of deformation of cells and microrelief of cellular surface of bacteria, and spatial structure of microorganism microfilms and extracellular polymer matrix in process of drying.
